An electrochemiluminescence (ECL) biosensor based on tRNA-loaded AuNPs was established for the detection of HIV-1. A novel biosensing strategy was applied, in which the metal free chemiluminescent ECL signal was mediated by tRNA-AuNPs bio-receptors with catalytic activity on tRNA-dependent recognition of HIV-1 RNA. The ECL signal-off system was observed to function through protection of the 1-aminonaphthalene-4-sulfonic acid (1,4-ANS) emitting molecules against quencher interference from the environment. The limit of detection (LOD) of the system was estimated to be as low as 10 pM, which is equivalent to 10 copies of HIV-1 RNA. The as-prepared system has also been used for sensitive determination of HIV-1 in clinical samples. The main components of the ECL biosensor, including tRNA-AuNPs bio-receptors, have been fully characterized. The novel biosensing strategy offers a powerful platform for the detection
